BRCA mutations have been known to be associated with breast cancer and ovarian cancer for more than two decades. Despite advances in care, for newly diagnosed patients the overall survival at 5 years has only marginally improved to 46% during the last 20 years. The two most important factors for the lack of improvement are a high rate of advanced disease at diagnosis and lack of new therapeutic options.
More recent studies have shown that BRCA mutations are also linked to many cases of advanced prostate cancer, as well as pancreatic cancer. Those prostate cancers that have spread or are more aggressive are more likely to be associated with a hereditary predisposition. In 2019, a study found that BRCA2 mutations are associated with poorer treatment responses and outcomes among men with aggressive prostate cancer (a 2014 study found that the 12-year prostate cancer-specific survival rate was 94.3% for men without a BRCA2 mutation but was only 61.8% for men with a mutation).
